APS3

YKS7, L000003174, YJL024C
Small subunit of the clathrin-associated adaptor complex AP-3; involved in vacuolar protein sorting; related to the sigma subunit of the mammalian clathrin AP-3 complex; suppressor of loss of casein kinase 1 function; protein abundance increases in response to DNA replication stress

VPS5

GRD2, PEP10, VPT5, YOR29-20, L000002920, YOR069W
Nexin-1 homolog; required for localizing membrane proteins from a prevacuolar/late endosomal compartment back to late Golgi; structural component of retromer membrane coat complex; forms a retromer subcomplex with Vps17p; required for recruiting the retromer complex to the endosome membranes; VPS5 has a paralog, YKR078W, that arose from the whole genome duplication

Negative Genetic

Mutations/deletions in separate genes, each of which alone causes a minimal phenotype, but when combined in the same cell results in a more severe fitness defect or lethality under a given condition. This term is reserved for high or low throughput studies with scores.

We generated a global genetic interaction network for Saccharomyces cerevisiae, constructing more than 23 million double mutants, identifying about 550,000 negative and about 350,000 positive genetic interactions. This comprehensive network maps genetic interactions for essential gene pairs, highlighting essential genes as densely connected hubs. Genetic interaction profiles enabled assembly of a hierarchical model of cell function, including modules corresponding to ... [more]
