dut-1 encodes a deoxyuridine triphosphate nucleotidohydrolase (dUTPase); by homology, DUT-1 is predicted to catalyze the hydrolysis of dUTP into dUMP and pyrophosphate, thus providing the dUMP substrate for de novo dTMP synthesis; disruption of DUT-1 activity leads to incorporation of uracil into DNA which can be removed by the base excision repair pathway, but eventually gives rise to apoptosis and lethality owing to exceeding the repair capacity of the organism.
