Affinity Capture-Western
An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

CUEDC1 is a primary target of ER? essential for the growth of breast cancer cells.
Breast cancer is the most prevalent type of malignancy in women with ?1.7 million new cases diagnosed annually, of which the majority express ER? (ESR1), a ligand-dependent transcription factor. Genome-wide chromatin binding maps suggest that ER? may control the expression of thousands of genes, posing a great challenge in identifying functional targets. Recently, we developed a CRISPR-Cas9 functional genetic screening ... [more]
