Affinity Capture-MS
An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner is identified by mass spectrometric methods.

Multilevel proteomics reveals host perturbations by SARS-CoV-2 and SARS-CoV.
The global emergence of SARS-CoV-2 urgently requires an in-depth understanding of molecular functions of viral proteins and their interactions with the host proteome. Several individual omics studies have extended our knowledge of COVID-19 pathophysiology1-10. Integration of such datasets to obtain a holistic view of virus-host interactions and to define the pathogenic properties of SARS-CoV-2 is limited by the heterogeneity of ... [more]

Additional Notes
- Affinity capture-MS was carried out using HA-tagged viral proteins as baits in human A549 lung carcinoma cells. Significant interactions were identified as those where the prey protein was at least 4 times enriched against the background (median log2 score > 2) with a p-value <= 1E-3.
