Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

An initial biochemical and cell biological characterization of the mammalian homologue of a central plant developmental switch, COP1.

Yi C, Wang H, Wei N, Deng XW

Constitutive photomorphogenic 1 (COP1) has been defined as a central regulator of photomorphogenic development in plants, which targets key transcription factors for proteasome-dependent degradation. Although COP1 mammalian homologue has been previously reported, its function and distribution in animal kingdom are not known. Here we report the characterization of full-length human and mouse COP1 cDNAs and the genomic structures of the COP1 ...
