RRP6

exosome nuclease subunit RRP6, L000003540, YOR001W
Nuclear exosome exonuclease component; has 3'-5' exonuclease activity that is regulated by Lrp1p; involved in RNA processing, maturation, surveillance, degradation, tethering, and export; role in sn/snoRNAs precursor degradation; forms a stable heterodimer with Lrp1p; has similarity to E. coli RNase D and to human PM-Sc1 100 (EXOSC10); mutant displays reduced transcription elongation in the G-less-based
Saccharomyces cerevisiae (S288c)

SHE11, RNA-binding protein YRA1, L000002959, L000002873, YDR381W
Nuclear polyadenylated RNA-binding protein; required for export of poly(A)+ mRNA from the nucleus; proposed to couple mRNA export with 3' end processing via its interactions with Mex67p and Pcf11p; interacts with DBP2; inhibits the helicase activity of Dbp2; functionally redundant with Yra2p, another REF family member

Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

Stable mRNP formation and export require cotranscriptional recruitment of the mRNA export factors Yra1p and Sub2p by Hpr1p.

Zenklusen D, Vinciguerra P, Wyss JC, Stutz F

Yra1p/REF participates in mRNA export by recruiting the export receptor Mex67p to messenger ribonucleoprotein (mRNP) complexes. Yra1p also binds Sub2p, a DEAD box ATPase/RNA helicase implicated in splicing and required for mRNA export. We identified genetic and physical interactions between Yra1p, Sub2p, and Hpr1p, a protein involved in transcription elongation whose deletion leads to poly(A)(+) RNA accumulation in the nucleus. ...
