BRF1

PCF4, TDS4, transcription factor TFIIIB subunit BRF1, TFIIIB70, L000000193, YGR246C
TFIIIB B-related factor; one of three subunits of RNA polymerase III transcription initiation factor TFIIIB, binds TFIIIC and TBP and recruits RNA pol III to promoters, amino-terminal half is homologous to TFIIB
Saccharomyces cerevisiae (S288c)

MAF1

RNA polymerase III-inhibiting protein MAF1, L000002614, YDR005C
Highly conserved negative regulator of RNA polymerase III; involved in tRNA processing and stability; inhibits tRNA degradation via rapid tRNA decay (RTD) pathway; binds N-terminal domain of Rpc160p subunit of Pol III to prevent closed-complex formation; localization and activity are regulated by phosphorylation, mediated by TORC1, protein kinase A, and Sch9p; localizes to cytoplasm during vegetative growth and translocates to nucleus and nucleolus under stress conditions

Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

Two steps in Maf1-dependent repression of transcription by RNA polymerase III.

Desai N, Lee J, Upadhya R, Chu Y, Moir RD, Willis IM

In Saccharomyces cerevisiae, Maf1 is essential for mediating the repression of transcription by RNA polymerase (pol) III in response to diverse cellular conditions. These conditions activate distinct signaling pathways that converge at or above Maf1. Thus, Maf1-dependent repression is thought to involve a common set of downstream inhibitory effects on the pol III machinery. Here we provide support for this ... [more]
