BAIT

MLP2

S000007510, YIL149C
Myosin-like protein associated with the nuclear envelope; nuclear basket protein that connects the nuclear pore complex with the nuclear interior; involved in the Tel1p pathway that controls telomere length; MLP2 has a paralog, MLP1, that arose from the whole genome duplication
Saccharomyces cerevisiae (S288c)

KAP95

RSL1, L000002781, L000002885, YLR347C
Karyopherin beta; forms a complex with Srp1p/Kap60p; interacts with nucleoporins to mediate nuclear import of NLS-containing cargo proteins via the nuclear pore complex; regulates PC biosynthesis; GDP-to-GTP exchange factor for Gsp1p
Saccharomyces cerevisiae (S288c)

Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

Mlp2p, a component of nuclear pore attached intranuclear filaments, associates with nic96p.

A fraction of the yeast nucleoporin Nic96p is localized at the terminal ring of the nuclear basket. When Nic96p was affinity purified from glutaraldehyde-treated spheroplasts, it was found to be associated with Mlp2p. Mlp2p, together with Mlp1p, are the yeast Tpr homologues, which form the nuclear pore-attached intranuclear filaments (Strambio-de-Castillia, C., Blobel, G., and Rout, M. P. (1999) J. Cell ... Mutations/deletions in separate genes, each of which alone causes a minimal phenotype, but when combined in the same cell results in a more severe fitness defect or lethality under a given condition. This term is reserved for high or low throughput studies with scores.
