Phenotypic Suppression
A genetic interaction is inferred when mutation or over expression of one gene results in suppression of any phenotype (other than lethality/growth defect) associated with mutation or over expression of another gene.

gp78 functions downstream of Hrd1 to promote degradation of misfolded proteins of the endoplasmic reticulum.
Eukaryotic cells eliminate misfolded proteins from the endoplasmic reticulum (ER) via a conserved process termed ER-associated degradation (ERAD). Central regulators of the ERAD system are membrane-bound ubiquitin ligases, which are thought to channel misfolded proteins through the ER membrane during retrotranslocation. Hrd1 and gp78 are mammalian ubiquitin ligases homologous to Hrd1p, an ubiquitin ligase essential for ERAD in Saccharomyces cerevisiae. ... [more]
Throughput
- Low Throughput
Ontology Terms
- phenotype: abnormal protein level (MP:0008469)
Additional Notes
- CRISPR/Cas9
- the accumulation of ubiquitinated MHC 1-147 and retrotranslocated deglycosylated heavy chain molecules seen in a Bag6 or gp78 knockdown condition was reversed upon depletion of Hrd1 as measured by the split-GFP assay

AMFR SYVN1 | Affinity Capture-Western Affinity Capture-Western An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins. | Low | - | BioGRID | - | |
