ribonucleotide reductase inhibiting protein SML1, L000004580, YML058W
Ribonucleotide reductase inhibitor; involved in regulating dNTP production; regulated by Mec1p and Rad53p during DNA damage and S phase; SML1 has a paralog, DIF1, that arose from the whole genome duplication

CRT7, RIR1, SDS12, ribonucleotide-diphosphate reductase subunit RNR1, L000001655, YER070W
Major isoform of large subunit of ribonucleotide-diphosphate reductase; the RNR complex catalyzes rate-limiting step in dNTP synthesis, regulated by DNA replication and DNA damage checkpoint pathways via localization of small subunits; relative distribution to the nucleus increases upon DNA replication stress; RNR1 has a paralog, RNR3, that arose from the whole genome duplication

Inhibition of yeast ribonucleotide reductase by Sml1 depends on the allosteric state of the enzyme.

Sml1 is an intrinsically disordered protein inhibitor of Saccharomyces cerevisiae ribonucleotide reductase (ScRR1), but its inhibition is poorly understood. RR reduces ribonucleoside diphosphates to their deoxy forms, and balances the nucleotide pool. Multiple turnover kinetics show that Sml1 inhibition of dGTP/ADP and ATP/CDP bound ScRR follows a mixed inhibition mechanism. However, Sml1 cooperatively binds to the ES complex in the ... [more]

Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.
