WSS1

YHR134W
Metalloprotease involved in DNA repair, removes DNA-protein crosslinks at stalled replication forks during replication of damaged DNA; sumoylated protein localizing to the nuclear periphery of mother cells; localizes to a single spot on the nuclear periphery of mother cells but not daughters; interacts genetically with SMT3; activated by DNA binding

RPL17A

RPL17, ribosomal 60S subunit protein L17A, L22, YL17, L20A, L17A, L000001715, YKL180W
Ribosomal 60S subunit protein L17A; copurifies with the Dam1 complex (aka DASH complex); homologous to mammalian ribosomal protein L17 and bacterial L22; RPL17A has a paralog, RPL17B, that arose from the whole genome duplication; protein abundance increases in response to DNA replication stress

Affinity Capture-MS

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner is identified by mass spectrometric methods.

Sumoylation during genotoxic stress regulates the composition of DNA repair complexes. The yeast metalloprotease Wss1 clears chromatin-bound sumoylated proteins. Wss1 and its mammalian analog, DVC1/Spartan, belong to minigluzincins family of proteases. Wss1 proteolytic activity is regulated by a cysteine switch mechanism activated by chemical stress and/or DNA binding. Wss1 is required for cell survival following UV irradiation, the smt3-331 mutation ... [more]

Mutations/deletions in separate genes, each of which alone causes a minimal phenotype, but when combined in the same cell results in a more severe fitness defect or lethality under a given condition. This term is reserved for high or low throughput studies with scores.
