SNP1

U1-70K, L000001954, YIL061C
Component of U1 snRNP required for mRNA splicing via spliceosome; substrate of the arginine methyltransferase Hmt1p; may interact with poly(A) polymerase to regulate polyadenylation; homolog of human U1 70K protein; protein abundance increases in response to DNA replication stress

YPL157W
Trimethyl guanosine synthase, conserved nucleolar methyl transferase; converts the m(7)G cap structure of snRNAs, snoRNAs, and telomerase TLC1 RNA to m(2,2,7)G; also required for nucleolar assembly and splicing of meiotic pre-mRNAs; interacts with Swm2p, which may confer substrate specificity on Tgs1p

Synthetic Growth Defect

A genetic interaction is inferred when mutations in separate genes, each of which alone causes a minimal phenotype, result in a significant growth defect under a given condition when combined in the same cell.

Two Routes to Genetic Suppression of RNA Trimethylguanosine Cap Deficiency via C-Terminal Truncation of U1 snRNP Subunit Snp1 or Overexpression of RNA Polymerase Subunit Rpo26.

Qiu ZR, Schwer B, Shuman S

The trimethylguanosine (TMG) caps of small nuclear (sn) RNAs are synthesized by the enzyme Tgs1 via sequential methyl additions to the N2 atom of the m(7)G cap. Whereas TMG caps are inessential for Saccharomyces cerevisiae vegetative growth at 25° to 37°, tgs1∆ cells that lack TMG caps fail to thrive at 18°. The cold-sensitive defect correlates with ectopic stoichiometric association ... [more]
