UBC7

DER2, QRI8, E2 ubiquitin-conjugating protein UBC7, L000001552, YMR022W
Ubiquitin conjugating enzyme; involved in the ER-associated protein degradation pathway; requires Cue1p for recruitment to the ER membrane; proposed to be involved in chromatin assembly
Saccharomyces cerevisiae (S288c)
PREY

HAC1

ERN4, IRE15, transcription factor HAC1, L000002611, YFL031W
Basic leucine zipper (bZIP) transcription factor (ATF/CREB1 homolog); regulates the unfolded protein response, via UPRE binding, and membrane biogenesis; ER stress-induced splicing pathway facilitates efficient Hac1p synthesis; protein abundance increases in response to DNA replication stress
Saccharomyces cerevisiae (S288c)

Synthetic Lethality

A genetic interaction is inferred when mutations or deletions in separate genes, each of which alone causes a minimal phenotype, result in lethality when combined in the same cell under a given condition.

A regulatory link between ER-associated protein degradation and the unfolded-protein response.

Ubiquitin conjugation during endoplasmic-reticulum-associated degradation (ERAD) depends on the activity of Ubc7. Here we show that Ubc1 acts as a further ubiquitin-conjugating enzyme in this pathway. Absence of both enzymes results in marked stabilization of an ERAD substrate and induction of the unfolded-protein response (UPR). Furthermore, basic ERAD activity is sufficient to eliminate unfolded proteins under normal conditions. However, when ... [more]
