BAIT

NFS1

SPL1, L000001240, YCL017C
Cysteine desulfurase; involved in iron-sulfur cluster (Fe/S) biogenesis and in thio-modification of mitochondrial and cytoplasmic tRNAs; essential protein located predominantly in mitochondria
Saccharomyces cerevisiae (S288c)
PREY

ATG5

APG5, L000003303, YPL149W
Conserved protein involved in autophagy and the Cvt pathway; undergoes conjugation with Atg12p to form a complex involved in Atg8p lipidation; Atg5p-Atg12p conjugate also forms a complex with Atg16p; the Atg5-Atg12/Atg16 complex binds to membranes and is essential for autophagosome formation; also involved in methionine restriction extension of chronological lifespan in an autophagy-dependent manner
Saccharomyces cerevisiae (S288c)

Negative Genetic

Mutations/deletions in separate genes, each of which alone causes a minimal phenotype, but when combined in the same cell results in a more severe fitness defect or lethality under a given condition. This term is reserved for high or low throughput studies with scores.

We generated a global genetic interaction network for Saccharomyces cerevisiae, constructing more than 23 million double mutants, identifying about 550,000 negative and about 350,000 positive genetic interactions. This comprehensive network maps genetic interactions for essential gene pairs, highlighting essential genes as densely connected hubs. Genetic interaction profiles enabled assembly of a hierarchical model of cell function, including modules corresponding to ... [more]
