Component of the exomer complex; exomer also contains Csh6p, Bch1p, Bch2p, and Bud7p and is involved in export of selected proteins, such as chitin synthase Chs3p, from the Golgi to the plasma membrane; Chs5p is the only protein with a BRCT domain that is not localized to the nucleus

Clathrin light chain; subunit of the major coat protein involved in intracellular protein transport and endocytosis; regulates endocytic progression; thought to regulate clathrin function; the clathrin triskelion is a trimeric molecule composed of three heavy chains that radiate from a vertex and three light chains which bind noncovalently near the vertex of the triskelion

Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

Chs5/6 complex: a multiprotein complex that interacts with and conveys chitin synthase III from the trans-Golgi network to the cell surface.

Sanchatjate S, Schekman R

In Saccharomyces cerevisiae, the polysaccharide chitin is deposited at the mother bud junction by an integral membrane enzyme, chitin synthase 3 (Chs3p). The traffic of Chs3p to the cell surface from the trans-Golgi network (TGN) depends on two proteins, Chs5p and Chs6p, which sort selected cargo proteins into secretory vesicles. We have found that Chs5p forms a large higher-order complex ... [more]

Mutations/deletions in separate genes, each of which alone causes a minimal phenotype, but when combined in the same cell results in a more severe fitness defect or lethality under a given condition. This term is reserved for high or low throughput studies with scores.
