DNA-directed RNA polymerase core subunit RPB5, ABC27, L000001590, L000001679, YBR154C
RNA polymerase subunit ABC27; common to RNA polymerases I, II, and III; contacts DNA and affects transactivation
Saccharomyces cerevisiae (S288c)

RSC4

L000004026, YKR008W
Component of the RSC chromatin remodeling complex; found in close proximity to nucleosomal DNA; displaced from the surface of nucleosomal DNA after chromatin remodeling; acetylated (K25) by Gcn5p, altering replication stress tolerance; contains tandem bromodomains that recognize histone H3 acetylated on K14 (H3K14ac) by Gcn5p

RSC is an essential, multisubunit chromatin remodeling complex. We show here that the Rsc4 subunit of RSC interacted via its C terminus with Rpb5, a conserved subunit shared by all three nuclear RNA polymerases (Pol). Furthermore, the RSC complex coimmunoprecipitated with all three RNA polymerases. Mutations in the C terminus of Rsc4 conferred a thermosensitive phenotype and the loss of ... [more]

Reconstituted Complex

An interaction is inferred between proteins in vitro. This can include proteins in recombinant form or proteins isolated directly from cells with recombinant or purified bait. For example, GST pull-down assays where a GST-tagged protein is first isolated and then used to fish interactors from cell lysates are considered reconstituted complexes (e.g. PUBMED: 14657240, Fig. 4A or PUBMED: 14761940, Fig. 5). This can also include gel-shifts, surface plasmon resonance, isothermal titration calorimetry (ITC) and bio-layer interferometry (BLI) experiments. The bait-hit directionality may not be clear for 2 interacting proteins. In these cases the directionality is up to the discretion of the curator.

Synthetic Lethality

A genetic interaction is inferred when mutations or deletions in separate genes, each of which alone causes a minimal phenotype, result in lethality when combined in the same cell under a given condition.
