GAC1

protein phosphatase regulator GAC1, L000000657, YOR178C
Regulatory subunit for Glc7p type-1 protein phosphatase (PP1); tethers Glc7p to Gsy2p glycogen synthase, binds Hsf1p heat shock transcription factor, required for induction of some HSF-regulated genes under heat shock; GAC1 has a paralog, PIG1, that arose from the whole genome duplication
Saccharomyces cerevisiae (S288c)

YTA4, L000001200, L000002558, YGR028W
Highly-conserved N-terminally anchored AAA-ATPase; distributed in the mitochondrial outer membrane and peroxisomes; involved in mitochondrial protein sorting; functions as an extraction engine in local organelle surveillance to remove and initiate degradation of mistargeted proteins, ensuring fidelity of organelle-specific localization of tail-anchored proteins; contains an N-terminal transmembrane domain and C-terminal cytoplasmic ATPase domain

Negative Genetic

Mutations/deletions in separate genes, each of which alone causes a minimal phenotype, but when combined in the same cell results in a more severe fitness defect or lethality under a given condition. This term is reserved for high or low throughput studies with scores.

We generated a global genetic interaction network for Saccharomyces cerevisiae, constructing more than 23 million double mutants, identifying about 550,000 negative and about 350,000 positive genetic interactions. This comprehensive network maps genetic interactions for essential gene pairs, highlighting essential genes as densely connected hubs. Genetic interaction profiles enabled assembly of a hierarchical model of cell function, including modules corresponding to ... [more]
