ORC2

RRR1, SIR5, origin recognition complex subunit 2, L000001776, YBR060C
Subunit of the origin recognition complex (ORC); ORC directs DNA replication by binding to replication origins and is also involved in transcriptional silencing; interacts with Spp1p and with trimethylated histone H3; phosphorylated by Cdc28p
Saccharomyces cerevisiae (S288c)

MCM7

CDC47, MCM DNA helicase complex subunit MCM7, L000002760, YBR202W
Component of the Mcm2-7 hexameric helicase complex; MCM2-7 primes origins of DNA replication in G1 and becomes an active ATP-dependent helicase that promotes DNA melting and elongation in S-phase; forms an Mcm4p-6p-7p subcomplex
Saccharomyces cerevisiae (S288c)

To initiate DNA replication, the origin recognition complex (ORC) and Cdc6 load an Mcm2-7 double hexamer onto DNA. Without ATP hydrolysis, ORC-Cdc6 recruits one Cdt1-bound Mcm2-7 hexamer, thus forming an ORC-Cdc6-Cdt1-Mcm2-7 (OCCM) helicase-loading intermediate. Here we report a 3.9-A structure of Saccharomyces cerevisiae OCCM on DNA. Flexible Mcm2-7 winged-helix domains (WHDs) engage ORC-Cdc6. A three-domain Cdt1 configuration embraces Mcm2, Mcm4, ... Synthetic Lethality

A genetic interaction is inferred when mutations or deletions in separate genes, each of which alone causes a minimal phenotype, result in lethality when combined in the same cell under a given condition.
