Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

Silencing of fused toes homolog enhances cisplatin sensitivity in cervical cancer cells by inhibiting epidermal growth factor receptor-mediated repair of DNA damage.

Subramanian PD, An Z, Yu JR, Park WY

Overexpression of epidermal growth factor receptor (EGFR) is related to chemo-/radioresistance and poor prognosis in many cancers. EGFR is activated by cisplatin, and this may lead to resistance to this drug. Fused toes homolog (FTS) is an E2 variant that lacks the active cysteine residue required for ubiquitin transfer. Previously, we reported that FTS interacts with EGFR and activates DNA-dependent ... [more]
