GPI18

FMP44, YBR004C
Functional ortholog of human PIG-V; PIG-V is a mannosyltransferase that transfers the second mannose in glycosylphosphatidylinositol biosynthesis; the authentic, non-tagged protein was localized to mitochondria
Saccharomyces cerevisiae (S288c)

PGA1

YNL158W
Essential component of GPI-mannosyltransferase II; complex is responsible for second mannose addition to GPI precursors as a partner of Gpi18p; required for maturation of Gas1p and Pho8p; has synthetic genetic interactions with secretory pathway genes

Dosage Rescue

A genetic interaction is inferred when over expression or increased dosage of one gene rescues the lethality or growth defect of a strain that is mutated or deleted for another gene.

Publication

Pga1 is an essential component of Glycosylphosphatidylinositol-mannosyltransferase II of Saccharomyces cerevisiae.

The Saccharomyces cerevisiae essential gene YNL158w/PGA1 encodes an endoplasmic reticulum (ER)-localized membrane protein. We constructed temperature-sensitive alleles of PGA1 by error-prone polymerase chain reaction mutagenesis to explore its biological role. Pulse-chase experiments revealed that the pga1(ts) mutants accumulated the ER-form precursor of Gas1 protein at the restrictive temperature. Transport of invertase and carboxypeptidase Y were not affected. Triton X-114 phase ... [more]

Mutations/deletions in separate genes, each of which alone causes a minimal phenotype, but when combined in the same cell results in a more severe fitness defect or lethality under a given condition. This term is reserved for high or low throughput studies with scores.
