- hits were identified in proximity labeling experiments in which each hit protein was fused to biotin acceptor peptide (AP) and the bait protein WWP1 was fused to wild type BirA (not the mutant BirA version used in BioID experiments); close proximity between the bait and hit protein, as would be expected when an E3 ubiquitin ligase such as WWP1 binds its substrates, then leads to AP biotinylation, allowing for affinity-based hit purification and identification by mass spectrometry
