Proximity Label-MS
An interaction is inferred when a bait-enzyme fusion protein selectively modifies a vicinal protein with a diffusible reactive product, followed by affinity capture of the modified protein and identification by mass spectrometric methods.

β-dystroglycan is regulated by a balance between WWP1-mediated degradation and protection from WWP1 by dystrophin and utrophin.
Dystroglycan is a ubiquitous membrane protein that functions as a mechanical connection between the extracellular matrix and cytoskeleton. In skeletal muscle, dystroglycan plays an indispensable role in regulating muscle regeneration; a malfunction in dystroglycan is associated with muscular dystrophy. The regulation of dystroglycan stability is poorly understood. Here, we report that WWP1, a member of NEDD4 E3 ubiquitin ligase family, ... [more]

Additional Notes
- hits were identified in proximity labeling experiments in which each hit protein was fused to biotin acceptor peptide (AP) and the bait protein WWP1 was fused to wild type BirA (not the mutant BirA version used in BioID experiments); close proximity between the bait and hit protein, as would be expected when an E3 ubiquitin ligase such as WWP1 binds its substrates, then leads to AP biotinylation, allowing for affinity-based hit purification and identification by mass spectrometry
