Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

STAT5A interacts with and is phosphorylated upon activation of the mu-opioid receptor.

Mazarakou G, Georgoussi Z

Signal Transducers and Activators of Transcription (STATs) are transcription factors shown to be activated by G protein-coupled receptors. In the present study, we demonstrate that acute morphine or [D-Ala(2),N-Me-Phe(4),Gly(5)-ol]enkephalin (DAMGO) exposure of COS-7 cells transiently transfected with the micro-opioid receptor and STAT5A, leads to receptor-dependent tyrosine phosphorylation of STAT5A. Activation of HEK293 cells, stably expressing the micro-opioid receptor with micro-opioid ... [more]
