Molecular interaction of NMDA receptor subunit NR3A with protein phosphatase 2A.

Ma OK, Sucher NJ

The catalytic subunit of the serine-threonine protein phosphatase 2A (PP2A) was previously found to bind to the carboxyl domain of NMDA receptor (NMDAR) subunit NR3A. We now report that NR3A constitutively associates with the PP2A holoenzyme, but not the core enzyme in rat brain synaptic plasma membranes. We also identified critical amino acids in NR3A required for binding to PP2A. ... [more]

Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.
