Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

YAP-dependent ubiquitination and degradation of ?-catenin mediates inhibition of Wnt signalling induced by Physalin F in colorectal cancer.

Chen C, Zhu D, Zhang H, Han C, Xue G, Zhu T, Luo J, Kong L

Aberrant activation of Wnt/?-catenin signalling is critical in the progression of human cancers, especially colorectal cancer (CRC). Therefore, inhibition of Wnt/?-catenin signalling is a significant potential target for CRC therapy. Here, we identified for the first time that Physalin F (PF), a steroid derivative isolated from Physalis angulate, acts as an antagonist of Wnt/?-catenin signalling. In vitro, PF decreased Wnt3a-induced ... [more]
