Affinity Capture-Western
An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

USP15 potentiates NF-?B activation by differentially stabilizing TAB2 and TAB3.
Tumor necrosis factor ? (TNF?)- and interleukin 1? (IL-1?)-induced nuclear factor-?B (NF-?B) activation play key roles in inflammation, immunity, and cancer development. Here, we identified one of the deubiquitinating enzymes (DUBs), ubiquitin-specific protease 15 (USP15), as a positive regulator in both TNF?- and IL-1?-induced NF-?B activation. Overexpression of USP15 potentiated TNF?- or IL-1?-triggered NF-?B activation and downstream gene transcription, whereas ... [more]
