Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

The Deubiquitinating Enzyme USP20 Regulates the TNF?-Induced NF-?B Signaling Pathway through Stabilization of p62.

Ha J, Kim M, Seo D, Park JS, Lee J, Lee J, Park SH

p62/sequestosome-1 is a scaffolding protein involved in diverse cellular processes such as autophagy, oxidative stress, cell survival and death. It has been identified to interact with atypical protein kinase Cs (aPKCs), linking these kinases to NF-?B activation by tumor necrosis factor ? (TNF?). The diverse functions of p62 are regulated through post-translational modifications of several domains within p62. Among the ... [more]
