MOT1

BUR3, END10, LPF4, BTAF1, L000001136, L000004559, S000029158, YPL082C
Essential protein involved in regulation of transcription; removes Spt15p (TBP) from DNA via its C-terminal ATPase activity; may have a role in ensuring that soluble TBP is available to bind TATA-less promoters; forms a complex with TBP that binds TATA DNA with high affinity but with altered specificity; the Mot1p-Spt15p-DNA ternary complex contains unbent DNA
Saccharomyces cerevisiae (S288c)

POB3

FACT complex subunit POB3, L000003326, YML069W
Subunit of the heterodimeric FACT complex (Spt16p-Pob3p); FACT associates with chromatin via interaction with Nhp6Ap and Nhp6Bp, and reorganizes nucleosomes to facilitate access to DNA by RNA and DNA polymerases; protein abundance increases in response to DNA replication stress
Saccharomyces cerevisiae (S288c)

Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

A proteomics analysis of yeast Mot1p protein-protein associations: insights into mechanism.

Arnett DR, Jennings JL, Tabb DL, Link AJ, Weil PA

Yeast Mot1p, a member of the Snf2 ATPase family of proteins, is a transcriptional regulator that has the unusual ability to both repress and activate mRNA gene transcription. To identify interactions with other proteins that may assist Mot1p in its regulatory processes, Mot1p was purified from replicate yeast cell extracts, and Mot1p-associated proteins were identified by coupled multidimensional liquid chromatography ... Mutations/deletions in separate genes, each of which alone causes a minimal phenotype, but when combined in the same cell results in a more severe fitness defect or lethality under a given condition. This term is reserved for high or low throughput studies with scores.
