Affinity Capture-Western
An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

Inhibition of major histocompatibility complex-I antigen presentation by sarbecovirus ORF7a proteins.
Viruses employ a variety of strategies to escape or counteract immune responses, including depletion of cell surface major histocompatibility complex class I (MHC-I), that would ordinarily present viral peptides to CD8+ cytotoxic T cells. As part of a screen to elucidate biological activities associated with individual severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) viral proteins, we found that ORF7a reduced cell ... Additional Notes
- SARS-CoV-2 Orf7a disrupts the MHC-1 peptide loading complex. The PLC includes the transporter associated with antigen processing (TAP) proteins, TAP1 and TAP2, and also the chaperones/cofactors Tapasin, ERp57 and calreticulin.
