DBF4

DNA52, LSD7, protein serine/threonine kinase activating protein DBF4, L000000488, S000029149, L000000513, YDR052C
Regulatory subunit of Cdc7p-Dbf4p kinase complex; required for Cdc7p kinase activity and initiation of DNA replication; phosphorylates the Mcm2-7 family of proteins; cell cycle regulated; relative distribution to the nucleus increases upon DNA replication stress
Saccharomyces cerevisiae (S288c)
PREY

MCM5

BOB1, CDC46, MCM DNA helicase complex subunit MCM5, L000000279, YLR274W
Component of the Mcm2-7 hexameric helicase complex; MCM complex is important for priming origins of DNA replication in G1 and becomes an active ATP-dependent helicase that promotes DNA melting and elongation when activated by Cdc7p-Dbf4p in S-phase
Saccharomyces cerevisiae (S288c)

Reconstituted Complex

An interaction is detected between purified proteins in vitro.

Dbf4-Cdc7 phosphorylation of Mcm2 is required for cell growth.

Bruck I, Kaplan D

The Dbf4-Cdc7 kinase (DDK) is required for the activation of the origins of replication, and DDK phosphorylates Mcm2 in vitro. We find that budding yeast Cdc7 alone exists in solution as a weakly active multimer. Dbf4 forms a likely heterodimer with Cdc7, and this species phosphorylates Mcm2 with substantially higher specific activity. Dbf4 alone binds tightly to Mcm2, whereas Cdc7 ... [more]
