Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

Deubiquitinase USP8 increases ID1 stability and promotes esophageal squamous cell carcinoma tumorigenesis.

ID1 is a labile protein implicated in the development and progression of several malignant tumors, but the mechanisms regulating ID1 stability and function in human esophageal squamous cell carcinoma (ESCC) are largely unclear. In this study, we performed an immunoprecipitation assay to screen for deubiquitinases that may interact with ID1 and identified USP8 as a novel deubiquitinase for ID1. USP8 ... [more]
