SWI4

ART1, SBF complex DNA-binding subunit SWI4, L000000124, L000002252, YER111C
DNA binding component of the SBF complex (Swi4p-Swi6p); a transcriptional activator that in concert with MBF (Mbp1-Swi6p) regulates late G1-specific transcription of targets including cyclins and genes required for DNA synthesis and repair; Slt2p-independent regulator of cold growth; acetylation at two sites, K1016 and K1066, regulates interaction with Swi6p
Saccharomyces cerevisiae (S288c)

POB3

FACT complex subunit POB3, L000003326, YML069W
Subunit of the heterodimeric FACT complex (Spt16p-Pob3p); FACT associates with chromatin via interaction with Nhp6Ap and Nhp6Bp, and reorganizes nucleosomes to facilitate access to DNA by RNA and DNA polymerases; protein abundance increases in response to DNA replication stress
Saccharomyces cerevisiae (S288c)

Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

FACT and Asf1 regulate nucleosome dynamics and coactivator binding at the HO promoter.

Takahata S, Yu Y, Stillman DJ

Transcriptional activators and coactivators overcome repression by chromatin, but regulation of chromatin disassembly and coactivator binding to promoters is poorly understood. Activation of the yeast HO gene follows the sequential binding of both sequence-specific DNA-binding proteins and coactivators during the cell cycle. Here, we show that the nucleosome disassembly occurs in waves both along the length of the promoter and ... [more]

Mutations/deletions in separate genes, each of which alone causes a minimal phenotype, but when combined in the same cell results in a more severe fitness defect or lethality under a given condition. This term is reserved for high or low throughput studies with scores.
