DPB11

protein kinase activating protein DPB11, L000003001, YJL090C
DNA replication initiation protein; loads DNA pol epsilon onto pre-replication complexes at origins; checkpoint sensor recruited to stalled replication forks by the checkpoint clamp complex where it activates Mec1p; along with Rfa1p, binds to ultrafine anaphase bridges in mitotic cells and prevents accumulation of chromatin bridges by stimulating the Mec1p kinase and suppressing homologous recombination; ortholog of human TopBP1; forms nuclear foci upon DNA replication stress
Saccharomyces cerevisiae (S288c)

MSA1

YOR29-17, YOR066W
Activator of G1-specific transcription factors MBF and SBF; involved in regulation of the timing of G1-specific gene transcription and cell cycle initiation; localization is cell-cycle dependent and regulated by Cdc28p phosphorylation; MSA1 has a paralog, MSA2, that arose from the whole genome duplication

Dosage Rescue

A genetic interaction is inferred when over expression or increased dosage of one gene rescues the lethality or growth defect of a strain that is mutated or deleted for another gene.

Identification of MSA1, a cell cycle-regulated, dosage suppressor of drc1/sld2 and dpb11 mutants.

Li JM, Tetzlaff MT, Elledge SJ

The Dpb11 and Drc1/Sld2 proteins form a complex that is critical for the initiation of DNA replication. In this study we identify MSA1 as a high copy suppressor of a drc1-1 mutant. MSA1 overproduction can also suppress the temperature sensitivity of dpb11-1 and pol2-12 mutants. Reciprocally, msa1 deletion exacerbates the mutant phenotypes of both drc1/sld2 and dpb11 mutants and msa1 ... [more]

Mutations/deletions in separate genes, each of which alone causes a minimal phenotype, but when combined in the same cell results in a more severe fitness defect or lethality under a given condition. This term is reserved for high or low throughput studies with scores.
