DED1

SPP81, DEAD-box ATP-dependent RNA helicase DED1, L000000500, YOR204W
ATP-dependent DEAD (Asp-Glu-Ala-Asp)-box RNA helicase; required for translation initiation of all yeast mRNAs; binds to mRNA cap-associated factors, and binding stimulates Ded1p RNA-dependent ATPase activity; mutation in human homolog DBY is associated with male infertility; human homolog DDX3X complements ded1 null mutation; DED1 has a paralog, DBP1, that arose from the whole genome duplication

Positive Genetic

Mutations/deletions in separate genes, each of which alone causes a minimal phenotype, but when combined in the same cell results in a less severe fitness defect than expected under a given condition. This term is reserved for high or low throughput studies with scores.

A synthetic genetic array screen for interactions with the RNA helicase DED1 during cell stress in budding yeast.

Carey SB, List HM, Siby A, Guerra P, Bolger TA

During cellular stress it is essential for cells to alter their gene expression to adapt and survive. Gene expression is regulated at multiple levels, but translation regulation is both a method for rapid changes to the proteome and, as one of the most energy-intensive cellular processes, a way to efficiently re-direct cellular resources during stress conditions. Despite this ideal positioning, ... [more]

A Protein-Fragment Complementation Assay (PCA) is a protein-protein interaction assay in which a bait protein is expressed as fusion to one of the either N- or C- terminal peptide fragments of a reporter protein and prey protein is expressed as fusion to the complementary N- or C- terminal fragment of the same reporter protein. Interaction of bait and prey proteins bring together complementary fragments, which can then fold into an active reporter, e.g. the split-ubiquitin assay.
