PRS2

ribose phosphate diphosphokinase subunit PRS2, L000001512, YER099C
5-phospho-ribosyl-1(alpha)-pyrophosphate synthetase, synthesizes PRPP; which is required for nucleotide, histidine, and tryptophan biosynthesis; one of five related enzymes, which are active as heteromultimeric complexes; PRS2 has a paralog, PRS4, that arose from the whole genome duplication

Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

Purification of proteasomes, proteasome subcomplexes, and proteasome-associated proteins from budding yeast.

Leggett DS, Glickman MH, Finley D

The proteasome is a highly complex, ATP-dependent protease, consisting of over 30 subunits, and dedicated mainly to the degradation of ubiquitin-protein conjugates. Proteasomes are evolutionarily conserved in the eukaryotic kingdom, and those of yeast are well suited to serve as a general model. We describe techniques for the purification of proteasomes from budding yeast in milligram amounts via conventional and ... [more]
