COX25, RRG10, YJL062W-A
Mitochondrial protein required for cytochrome c oxidase assembly; also involved in translational regulation of Cox1p and prevention of Cox1p aggregation before assembly; located in the mitochondrial inner membrane
Saccharomyces cerevisiae (S288c)

COX5A

cytochrome c oxidase subunit Va, L000000388, L000000387, YNL052W
Subunit Va of cytochrome c oxidase; cytochrome c oxidase is the terminal member of the mitochondrial inner membrane electron transport chain; Cox5Ap is predominantly expressed during aerobic growth while its isoform Vb (Cox5Bp) is expressed during anaerobic growth; COX5A has a paralog, COX5B, that arose from the whole genome duplication

Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

Cox25 teams up with Mss51, Ssc1 and Cox14 to regulate mitochondrial cytochrome C oxidase subunit 1 expression and assembly in Saccharomyces cerevisiae.

Fontanesi F, Clemente P, Barrientos A

In the yeast Saccharomyces cerevisiae, mitochondrial cytochrome c oxidase (COX) biogenesis is translationally regulated. Mss51, a specific COX1 mRNA translational activator and Cox1 chaperone, drives the regulatory mechanism. During translation and posttranslationally, newly synthesized Cox1 physically interacts with a complex of proteins involving Ssc1, Mss51 and Cox14, which eventually hand over Cox1 to the assembly pathway. This step is probably ... [more]
