HTZ1

HTA3, histone H2AZ, H2AZ, H2A.F/Z, L000003930, L000004094, YOL012C
Histone variant H2AZ; exchanged for histone H2A in nucleosomes by the SWR1 complex; involved in transcriptional regulation through prevention of the spread of silent heterochromatin; Htz1p-containing nucleosomes facilitate RNA Pol II passage by affecting correct assembly and modification status of RNA Pol II elongation complexes and by favoring efficient nucleosome remodeling
Saccharomyces cerevisiae (S288c)

HTA1

H2A1, SPT11, histone H2A, L000000827, YDR225W
Histone H2A; core histone protein required for chromatin assembly and chromosome function; one of two nearly identical subtypes (see also HTA2); DNA damage-dependent phosphorylation by Mec1p facilitates DNA repair; acetylated by Nat4p; N-terminally propionylated in vivo

Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

Stepwise histone replacement by SWR1 requires dual activation with histone H2A.Z and canonical nucleosome.

Histone variant H2A.Z-containing nucleosomes are incorporated at most eukaryotic promoters. This incorporation is mediated by the conserved SWR1 complex, which replaces histone H2A in canonical nucleosomes with H2A.Z in an ATP-dependent manner. Here, we show that promoter-proximal nucleosomes are highly heterogeneous for H2A.Z in Saccharomyces cerevisiae, with substantial representation of nucleosomes containing one, two, or zero H2A.Z molecules. SWR1-catalyzed H2A.Z ... [more]

Co-crystal Structure

Interaction directly demonstrated at the atomic level by X-ray crystallography. Also used for NMR or Electron Microscopy (EM) structures. If there is no obvious bait-hit directionality to the interaction involving 3 or more proteins, then the co-crystallized proteins should be listed as a complex.
