UBP3

BLM3, mRNA-binding ubiquitin-specific protease UBP3, L000002417, S000086717, YER151C
Ubiquitin-specific protease involved in transport and osmotic response; interacts with Bre5p to co-regulate anterograde and retrograde transport between the ER and Golgi; involved in transcription elongation in response to osmostress through phosphorylation at Ser695 by Hog1p; inhibitor of gene silencing; cleaves ubiquitin fusions but not polyubiquitin; also has mRNA binding activity; protein abundance increases in response to DNA replication stress; role in ribophagy

ANU1, COPII subunit SEC24, L000004360, YIL109C
Component of the Sec23p-Sec24p heterodimer of the COPII vesicle coat; required for cargo selection during vesicle formation in ER to Golgi transport; homologous to Sfb3p; SEC24 has a paralog, SFB2, that arose from the whole genome duplication

Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

Ubp3 requires a cofactor, Bre5, to specifically de-ubiquitinate the COPII protein, Sec23.

Ubiquitination is important for a broad array of cellular functions. Although reversal of this process, de-ubiquitination, most probably represents an important regulatory step contributing to cellular homeostasis, the specificity and properties of de-ubiquitination enzymes remain poorly understood. Here, we show that the Saccharomyces cerevisiae ubiquitin protease Ubp3 requires an additional protein, Bre5, to form an active de-ubiquitination complex that cleaves ... [more]
