MYO2
CDC66, myosin 2, L000001223, YOR326W
Type V myosin motor involved in actin-based transport of cargos; required for the polarized delivery of secretory vesicles, the vacuole, late Golgi elements, peroxisomes, and the mitotic spindle; MYO2 has a paralog, MYO4, that arose from the whole genome duplication

SAC1
RSD1, phosphatidylinositol-3-phosphatase SAC1, L000001790, YKL212W
Phosphatidylinositol phosphate (PtdInsP) phosphatase; involved in hydrolysis of PtdIns[4]P in the early and medial Golgi; regulated by interaction with Vps74p; ER localized transmembrane protein which cycles through the Golgi; involved in protein trafficking and processing, secretion, and cell wall maintenance; regulates sphingolipid biosynthesis through the modulation of PtdIns(4)P metabolism

Dosage Lethality
A genetic interaction is inferred when over expression or increased dosage of one gene causes lethality in a strain that is mutated or deleted for another gene.

PI4P and Rab inputs collaborate in myosin-V-dependent transport of secretory compartments in yeast.
Cell polarity involves transport of specific membranes and macromolecules at the right time to the right place. In budding yeast, secretory vesicles are transported by the myosin-V Myo2p to sites of cell growth. We show that phosphatidylinositol 4-phosphate (PI4P) is present in late secretory compartments and is critical for their association with, and transport by, Myo2p. Further, the trans-Golgi network ... [more]
