Affinity Capture-Western
An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

TNO1 is involved in salt tolerance and vacuolar trafficking in Arabidopsis thaliana.
The Arabidopsis t-SNARE (soluble N-ethylmaleimide-sensitive factor attachment protein receptor) SYP41 is involved in vesicle fusion at the trans-Golgi network (TGN) and interacts with AtVPS45, SYP61 and VTI12. These proteins are involved in diverse cellular processes, including vacuole biogenesis and stress tolerance. A previously uncharacterized protein, named TNO1, was identified by co-immunoprecipitation as a SYP41-interacting protein. TNO1 was found to localize ... [more]
