Affinity Capture-MS

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner is identified by mass spectrometric methods.

Dynamics of the putative RNA helicase Spb4 during ribosome assembly in Saccharomyces cerevisiae.

Garcia-Gomez JJ, Lebaron S, Froment C, Monsarrat B, Henry Y, de la Cruz J

Spb4 is a putative ATP-dependent RNA helicase required for proper processing of 27SB pre-rRNAs and therefore for 60S ribosomal subunit biogenesis. To define the timing of association of this protein with pre-ribosomal particles, we have studied the composition of complexes that copurify with TAP-tagged Spb4. These complexes contain mainly the 27SB pre-rRNAs and about 50 ribosome biogenesis proteins, primarily components ... [more]

Mutations/deletions in separate genes, each of which alone causes a minimal phenotype, but when combined in the same cell results in a more severe fitness defect or lethality under a given condition. This term is reserved for high or low throughput studies with scores.
