Vacuolar Fe2+/Mn2+ transporter; suppresses respiratory deficit of yfh1 mutants, which lack the ortholog of mammalian frataxin, by preventing mitochondrial iron accumulation; relative distribution to the vacuole decreases upon DNA replication stress

TYW1

YPL207W
Iron-sulfer protein required for synthesis of Wybutosine modified tRNA; Wybutosine is a modified guanosine found at the 3'-position adjacent to the anticodon of phenylalanine tRNA which supports reading frame maintenance by stabilizing codon-anticodon interactions; induction by Yap5p in response to iron provides protection from high iron toxicity; overexpression results in increased cellular iron

Synthetic Growth Defect

A genetic interaction is inferred when mutations in separate genes, each of which alone causes a minimal phenotype, result in a significant growth defect under a given condition when combined in the same cell.

Yap5-regulated transcription of TYW1 protects yeast from high iron toxicity.

Li L, Jia X, Ward DM, Kaplan J

The budding yeast Saccharomyces cerevisiae responds to high cytosolic iron by inducing Yap5-mediated transcription. We identified genes regulated by Yap5 in response to iron and show that one of the genes induced is TYW1, which encodes an iron-sulfur cluster enzyme that participates in the synthesis of Wybutosine (yW) modified t-RNA. Strains deleted for TYW1 do not show a phenotype in ... [more]
