Synthetic Rescue

A genetic interaction is inferred when mutations or deletions of one gene rescues the lethality or growth defect of a strain mutated or deleted for another gene.

The ubiquitin-like (UBX)-domain-containing protein Ubx2/ Ubxd8 regulates lipid droplet homeostasis.

Wang CW, Lee SC

Lipid droplets (LDs) are central organelles for maintaining lipid homeostasis. However, how cells control the size and number of LDs remains largely unknown. Herein, we report that Ubx2, a UBX-domain-containing protein involved in endoplasmic reticulum (ER)-associated degradation (ERAD), is crucial for LD maintenance. Ubx2 redistributes from ER to LDs when LDs start to form and enlarge during diauxic shift and ... [more]

Additional Notes

  • Ubxd8 expressed from the GPD1, but not UBX2 promoter, rescued the LD defect (Fig. 6B) and restored in ubx2 deletion mutant. In addition,Ubxd8 expression rescued the degradation defect of CPY*-HA and Deg1-GFP (Fig.3B), and the slow growth phenotype associated with ubx2 deletion mutant (Fig. 5C).
