ESS1

PIN1, PTF1, peptidylprolyl isomerase ESS1, L000000587, YJR017C
Peptidylprolyl-cis/trans-isomerase (PPIase); specific for phosphorylated serine and threonine residues N-terminal to proline; regulates phosphorylation of the RNAP II large subunit (Rpo21p) C-terminal domain (CTD); associates with phospho-Ser5 form of RNAP II in vivo; regulates phosphorylation of Ser7 within CTD; present along entire coding length of genes; represses initiation of CUTs; required for efficient termination of mRNA transcription and trimethylation of histone H3
Saccharomyces cerevisiae (S288c)

XRN1

DST2, KEM1, RAR5, SEP1, SKI1, chromatin-binding exonuclease XRN1, L000000891, L000001902, YGL173C
Evolutionarily-conserved 5'-3' exonuclease; component of cytoplasmic processing (P) bodies involved in mRNA decay; also enters the nucleus and positively regulates transcription initiation and elongation; plays a role in microtubule-mediated processes, filamentous growth, ribosomal RNA maturation, and telomere maintenance; activated by the scavenger decapping enzyme Dcs1p
Saccharomyces cerevisiae (S288c)

Synthetic Growth Defect

A genetic interaction is inferred when mutations in separate genes, each of which alone causes a minimal phenotype, result in a significant growth defect under a given condition when combined in the same cell.

Multiple Roles for the Ess1 Prolyl Isomerase in the RNA Polymerase II Transcription Cycle.

Ma Z, Atencio D, Barnes C, Defiglio H, Hanes SD

The Ess1 prolyl isomerase in Saccharomyces cerevisiae regulates RNA polymerase II (pol II) by isomerizing peptide bonds within the pol II CTD heptapeptide repeat (YSPTSPS). Ess1 preferentially targets the Ser5-Pro6 bond when Ser5 is phosphorylated. Conformational changes in the CTD induced by Ess1 control the recruitment of essential co-factors to the pol II complex, and may facilitate the ordered transition ... [more]
