Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

A novel HECT-type E3 ubiquitin protein ligase NEDL1 enhances the p53-mediated apoptotic cell death in its catalytic activity-independent manner.

Li Y, Ozaki T, Kikuchi H, Yamamoto H, Ohira M, Nakagawara A

NEDL1 (NEDD4-like ubiquitin protein ligase-1) is a newly identified HECT-type E3 ubiquitin protein ligase highly expressed in favorable neuroblastomas as compared with unfavorable ones. In this study, we found that NEDL1 cooperates with p53 to induce apoptosis. During cisplatin (CDDP)-mediated apoptosis in neuroblastoma SH-SY5Y cells, p53 was induced to accumulate in association with an increase in expression levels of NEDL1. ... [more]
