Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

Apolipoprotein J/clusterin in human erythrocytes is involved in the molecular process of defected material disposal during vesiculation.

Antonelou MH, Kriebardis AG, Stamoulis KE, Trougakos IP, Papassideri IS

We have showed that secretory Apolipoprotein J/Clusterin (sCLU) is down-regulated in senescent, stressed or diseased red blood cells (RBCs). It was hypothesized that sCLU loss relates to RBCs vesiculation, a mechanism that removes erythrocyte membrane patches containing defective or potentially harmful components. To investigate this issue we employed a combination of biochemical and microscopical approaches in freshly prepared RBCs or RBCs ...
