TRM7

YBR061C
2'-O-ribose methyltransferase; methylates the 2'-O-ribose of tRNA-Phe, tRNA-Trp, and tRNA-Leu at positions C32 and N34 of the tRNA anticodon loop; crucial biological role likely modification of tRNA-Phe; interacts with Trm732p and Rtt10p in 2'-O-methylation of C32 and N34 substrate tRNAs, respectively; functionally complemented by human FTSJ1

tRNA methyltransferase; two forms of protein are made by alternative translation starts; localizes to both nucleus and mitochondrion to produce modified base N2,N2-dimethylguanosine in tRNAs in both compartments; nuclear Trm1p is evenly distributed around inner membrane in WT, but mislocalizes as puncta near ER-nucleus junctions in spindle pole body (SPB) mutants; both Trm1p inner nuclear membrane targeting and maintenance depend upon SPB

Synthetic Lethality

A genetic interaction is inferred when mutations or deletions in separate genes, each of which alone causes a minimal phenotype, result in lethality when combined in the same cell under a given condition.

Post-transcriptional modification of the tRNA anticodon loop is critical for translation. Yeast Trm7 is required for 2'-O-methylation of C(32) and N(34) of tRNA(Phe), tRNA(Trp), and tRNA(Leu(UAA)) to form Cm(32) and Nm(34), and trm7-Δ mutants have severe growth and translation defects, but the reasons for these defects are not known. We show here that overproduction of tRNA(Phe) suppresses the growth defect ... [more]
