Affinity Capture-Western
An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

HectD1 E3 Ligase Modifies Adenomatous Polyposis Coli (APC) with Polyubiquitin to Promote the APC-Axin Interaction.
The adenomatous polyposis coli (APC) protein function as a negative regulator of the Wnt signaling pathway. In this capacity, APC forms a 'destruction complex' with Axin, CK1α and GSK3β to foster phosphorylation of the Wnt effector β-catenin earmarking it for K48-linked polyubiquitylation and proteasomal degradation. APC is conjugated with K63-linked ubiquitin chains when it is bound to Axin, but it ... [more]
