Regulation of ubiquitin transfer by XIAP, a dimeric RING E3 ligase.

Nakatani Y, Kleffmann T, Linke K, Condon SM, Hinds MG, Day CL

RING domains of E3 ligases promote transfer of ubiquitin (Ub) from the E2~Ub conjugate to target proteins. In many cases interaction of the E2~Ub conjugate with the RING domain requires its prior dimerization. Using cross-linking experiments we show that E2 conjugated ubiquitin contacts the RING homodimer interface of the Inhibitor of Apoptosis (IAP) proteins, XIAP and cIAP2. Structural and biochemical ... Co-crystal Structure

Interaction directly demonstrated at the atomic level by X-ray crystallography. Also used for NMR or Electron Microscopy (EM) structures. If there is no obvious bait-hit directionality to the interaction involving 3 or more proteins, then the co-crystallized proteins should be listed as a complex.
