Regulation of ubiquitin transfer by XIAP, a dimeric RING E3 ligase.

Nakatani Y, Kleffmann T, Linke K, Condon SM, Hinds MG, Day CL

RING domains of E3 ligases promote transfer of ubiquitin (Ub) from the E2~Ub conjugate to target proteins. In many cases interaction of the E2~Ub conjugate with the RING domain requires its prior dimerization. Using cross-linking experiments we show that E2 conjugated ubiquitin contacts the RING homodimer interface of the Inhibitor of Apoptosis (IAP) proteins, XIAP and cIAP2. Structural and biochemical ... Biochemical Activity

An interaction is inferred from the biochemical effect of one protein upon another, for example, GTP-GDP exchange activity or phosphorylation of a substrate by a kinase. The bait protein executes the activity on the substrate hit protein. A Modification value is recorded for interactions of this type with the possible values Phosphorylation, Ubiquitination, Sumoylation, Dephosphorylation, Methylation, Prenylation, Acetylation, Deubiquitination, Proteolytic Processing, Glucosylation, Nedd(Rub1)ylation, Deacetylation, No Modification, Demethylation.
